高效液相色谱-串联质谱法测定毛发中依托咪酯及依托咪酯酸

Determination of Etomidate and Etomidate acid in Human Hair by High Performance Liquid Chromotography Tandem Mass Spectroscopy

  • 摘要: 建立了一种高效液相色谱-串联质谱法(HPLC-MS/MS)测定毛发中依托咪酯和依托咪酯酸的方法,并应用于实际毛发样本的检测.将毛发洗净后剪碎、冷冻研磨,加入内标后以甲醇为溶剂超声提取,经氮吹浓缩,200μL甲醇定容后HPLC-MS/MS上机测定.依托咪酯和依托咪酯酸在线性范围(0.1~1 ng/mg)内相关系数分别为0.999、0.997,回收率在89%~113%,检出限分别为0.01 ng/mg和0.02 ng/mg,测定方法快速、灵敏、准确,可对毛发中两种药物进行同时定性和定量检测.

     

    Abstract: A method for screening Etomidate and Etomidate acid in human hair by high performance liquid chromotography tandem mass spectroscopy(HPLC-MS/MS)has been established and applied for the detection of real hair samples.The hair was washed,cut,frozen and ground,added to the internal standard,extracted ultrasonic with methanol as solvent,concentrated by nitrogen blowing.The concentrated extract was reconstituted to 100 μ L by methanol for HPLC-MS/MS analysis.The correlation coefficients of etomidate and etomidate acid in the linear range(0.1~lng/mg)were 0.999 and 0.997,the recoveries were 89%~113%,and the detection limits were 0.01ng/mg and 0.02ng/mg,respectively.The method was rapid,sensitive and accurate,and could be used for qualitative and quantitative detection of the two drugs in hair at the same time.
