Evaluation of Hypoglycemic and Hypolipidemic Effect of Hoematococcus pluvialis in Zebrafish

  • 摘要: 随着现代生活方式的变化,高糖高脂饮食结构成为导致糖尿病、肥胖、高脂血症和代谢综合症等疾病的主要因素之一。 针对这一问题,寻找安全有效的降糖降脂天然产物具有重要意义。雨生红球藻 (Hoematococcus pluvialis,HP) 作为含有丰 富虾青素 (Astaxanthin,ASX) 的天然资源,被认为可能具有调节血糖和血脂的潜力。本研究旨在通过斑马鱼 (Danio rerio, Zebrafish) 模型评估 HP 的降糖、降甘油三酯和降胆固醇效应,为其在代谢疾病治疗中的应用提供实验基础。本研究使用野 生型 AB 品系斑马鱼和黑色素等位基因突变 Albino 品系斑马鱼,通过高糖高脂饲料建立斑马鱼模型,HP 分别以 222、667 和 2000 μg/mL 的浓度进行处理,通过测定葡萄糖、甘油三酯、胆固醇等生化指标和观测斑马鱼尾静脉甘油三酯光密度和 胆固醇荧光强度等生物学特征,评估 HP 的降糖、降甘油三酯和降胆固醇效应。结果表明 HP 在 667 μg/mL 浓度下显著降 低了斑马鱼体内葡萄糖水平,在 2000 μg/mL 浓度下,除了显著降低葡萄糖水平外,还对甘油三酯和胆固醇水平产生了明 显的下调作用。HP 表现出对高糖高脂斑马鱼显著的降糖、降甘油三酯和降胆固醇作用,这为其作为潜在的保健品和药物 提供了初步的实验证据,尤其在维持血糖和血脂健康水平方面具有潜在应用前景。进一步研究有望揭示其具体的降糖降 脂机制和在临床应用中的可行性。

     

    Abstract: With the change of modern lifestyle, high glucose and high fat diet structure has become one of the main factors leading to diseases such as diabetes, obesity, hyperlipidemia and metabolic syndrome. To address this problem, it is of great significance to find safe and effective natural products lowering glucose and lipids. As a natural resource rich in astaxanthin, Hoematococcus pluvialis(HP) is considered to have the potential to regulate blood glucose and lipids. The aim of this study was to evaluate the effects of HP on lowering blood sugar, triglycerides, and cholesterol using the zebrafish model, and to provide an experimental basis for its application in the treatment of metabolic diseases. In this study, wild-type AB strain zebrafish and melanin allele mutant Albino strain zebrafish were used to establish a zebrafish model through high-glucose and high-fat diets, and HP was treated with concentrations of 222, 667 and 2000 μg/mL, respectively, to evaluate the hypoglycemic, triglyceride-lowering, and cholesterol-lowering effects of the HP through the measurement of glucose, triglyceride and cholesterol, as well as the biological characteristics such as the optical density of triglyceride and the fluorescence intensity of cholesterol in the caudal vein of the zebrafish. The glucose level of zebrafish was significantly reduced by HP at a concentration of 667 μg/mL. At a concentration of 2000 μg/mL, in addition to the significant reduction of glucose, it also produced significant down-regulation of triglyceride and cholesterol levels. The fact that HP exhibited significant glucose, triglyceride and cholesterol-lowering effects provides preliminary experimental evidence for the use of HP as potential nutraceuticals and medicines, with potential applications especially in maintaining healthy levels of blood glucose and blood lipids. Further studies are expected to reveal its specific hypoglycemic and hypolipidemic mechanisms and feasibility in clinical applications.
